Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of Caspase-11 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for Caspase-11 purchase    MBS741311 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Caspase 11 (Caspase-11) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 2.5-50ng/mL in biological research samples containing Caspase-11, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS741311 kit is based on Caspase-11 antibody-Caspase-11 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Caspase-11 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Caspase-11. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for Caspase-11 elisa kit

Principle of the Assay: Caspase-11 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Caspase-11 antibody and an Caspase-11-HRP conjugate. The assay sample and buffer are incubated together with Caspase-11-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Caspase-11 concentration since Caspase-11 from samples and Caspase-11-HRP conjugate compete for the anti-Caspase-11 antibody binding site. Since the number of sites is limited, as more sites are occupied by Caspase-11 from the sample, fewer sites are left to bind Caspase-11-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Caspase-11 concentration in each sample is interpolated from this standard curve.

NCBI Summary for Caspase-11    The protein encoded by this gene belongs to the membrane-associated guanylate kinase (MAGUK) family, a class of proteins that functions as molecular scaffolds for the assembly of multiprotein complexes at specialized regions of the plasma membrane. This protein is also a member of the CARD protein family, which is defined by carrying a characteristic caspase-associated recruitment domain (CARD). This protein has a domain structure similar to that of CARD14 protein. The CARD domains of both proteins have been shown to specifically interact with BCL10, a protein known to function as a positive regulator of cell apoptosis and NF-kappaB activation. When expressed in cells, this protein activated NF-kappaB and induced the phosphorylation of BCL10. [provided by RefSeq, Jul 2008]

UniProt Comments for Caspase-11    CARD11: a CARD (caspase recruitment domain) protein that belongs to the membrane-associated guanylate kinase (MAGUK) family. MAGUK proteins function as molecular scaffolds for the assembly of multiprotein complexes at specialized regions of the plasma membrane. Contains an N-terminal CARD domain, a central coiled-coil domain, and a C-terminal region containing a PDZ, an SH3, and a GUK. Its CARD domain associates with the CARD domain of BCL10. BCL10 activates NF-kappaB through the IkappaB kinase complex in response to upstream stimuli. Apparently activates NF-kappaB and induce the phosphorylation of BCL10. Detected in adult peripheral blood leukocytes, thymus, spleen and liver. Also found in promyelocytic leukemia HL-60 cells, chronic myelogenous leukemia K562 cells, Burkitt's lymphoma Raji cells and colorectal adenocarcinoma SW480 cells.
