Specificity    This assay has high sensitivity and excellent specificity for detection of GC. No significant cross-reactivity or interference between GCand analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between GC and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of GCG elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for GCG purchase    MBS7242321 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Glucagon (GC) (GCG) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing GCG, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS7242321 kit is based on GCG antibody-GCG antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GCG antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GCG. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses||This GC ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine GC. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: GC ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GC antibody and an GC-HRP conjugate. The assay sample and buffer are incubated together with GC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to theGC concentration since GC from samples and GC-HRP conjugate compete for the anti-GC antibody binding site. Since the number of sites is limited, as more sites are occupied by GC from the sample, fewer sites are left to bind GC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GC concentration in each sample is interpolated from this standard curve.

NCBI Summary for GCG    The protein encoded by this gene is actually a preproprotein that is cleaved into four distinct mature peptides. One of these, glucagon, is a pancreatic hormone that counteracts the glucose-lowering action of insulin by stimulating glycogenolysis and gluconeogenesis. Glucagon is a ligand for a specific G-protein linked receptor whose signalling pathway controls cell proliferation. Two of the other peptides are secreted from gut endocrine cells and promote nutrient absorption through distinct mechanisms. Finally, the fourth peptide is similar to glicentin, an active enteroglucagon. [provided by RefSeq, Jul 2008]

UniProt Comments for GCG    GCG: Glucagon plays a key role in glucose metabolism and homeostasis. Regulates blood glucose by increasing gluconeogenesis and decreasing glycolysis. A counterregulatory hormone of insulin, raises plasma glucose levels in response to insulin-induced hypoglycemia. Plays an important role in initiating and maintaining hyperglycemic conditions in diabetes. Glucagon release is stimulated by hypoglycemia and inhibited by hyperglycemia, insulin, and somatostatin. GLP-1 and GLP-2 are induced in response to nutrient ingestion. Glucagon is secreted in the A cells of the islets of Langerhans. GLP-1, GLP-2, oxyntomodulin and glicentin are secreted from enteroendocrine cells throughout the gastrointestinal tract. GLP1 and GLP2 are also secreted in selected neurons in the brain. Belongs to the glucagon family.

Research Articles on GCG    1. Data suggest that insulin secretion is down-regulated by aging and diabetes type 2 exacerbates this condition; secretion of adiponectin and glucagon-like peptide 1 (derived from glucagon) are also down-regulated.
