The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing LEP. The ELISA analytical biochemical technique of the MBS283334 kit is based on LEP antibody-LEP antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LEP antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LEP. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for LEP elisa kit

   Intended Uses: For the quantitative detection of Bovine Leptin (LEP) concentration in serum, plasma and other biological fluids.

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to LEP. Standards or samples are added to the appropriate microtiter plate wells with biotin-conjugated LEP. A competitive inhibition reaction is launched between LEP (Standards or samples) and biotin-conjugated LEP with the pre-coated antibody specific for LEP. The more amount of LEP in samples, the less antibody bound by biotin-conjugated LEP. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Substrate solution is added to the wells and the color develops in opposite to the amount of LEP in the sample. The color development is stopped and the intensity of the color is measured.

 NCBI Summary for LEP    This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development.
