 Searchable Terms for ACA-A purchase    MBS740100 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Activin A (ACA-A) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 50-1000pg/mL in biological research samples containing ACA-A, with an estimated sensitivity of 1.0pg/mL. The ELISA analytical biochemical technique of the MBS740100 kit is based on ACA-A antibody-ACA-A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ACA-A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ACA-A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

   Principle of the Assay: Activin-A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Activin-A antibody and an Activin-A-HRP conjugate. The assay sample and buffer are incubated together with Activin-A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Activin-A concentration since Activin-A from samples and Activin-A-HRP conjugate compete for the anti-Activin-A antibody binding site. Since the number of sites is limited, as more sites are occupied by Activin-A from the sample, fewer sites are left to bind Activin-A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Activin-A concentration in each sample is interpolated from this standard curve.

Intended Uses: This Activin-A ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine Activin-A. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

 NCBI Summary for ACA-A    The inhibin beta A subunit joins the alpha subunit to form a pituitary FSH secretion inhibitor. Inhibin has been shown to regulate gonadal stromal cell proliferation negatively and to have tumor-suppressor activity. In addition, serum levels of inhibin have been shown to reflect the size of granulosa-cell tumors and can therefore be used as a marker for primary as well as recurrent disease. Because expression in gonadal and various extragonadal tissues may vary severalfold in a tissue-specific fashion, it is proposed that inhibin may be both a growth/differentiation factor and a hormone. Furthermore, the beta A subunit forms a homodimer, activin A, and also joins with a beta B subunit to form a heterodimer, activin AB, both of which stimulate FSH secretion. Finally, it has been shown that the beta A subunit mRNA is identical to the erythroid differentiation factor subunit mRNA and that only one gene for this mRNA exists in the human genome.
