Product Description specifically for MS4A1 elisa kit

   Intended Uses: This MS4A1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine MS4A1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay||MS4A1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MS4A1 antibody and an MS4A1-HRP conjugate. The assay sample and buffer are incubated together with MS4A1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MS4A1 concentration since MS4A1 from samples and MS4A1-HRP conjugate compete for the anti-MS4A1 antibody binding site. Since the number of sites is limited, as more sites are occupied by MS4A1 from the sample, fewer sites are left to bind MS4A1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MS4A1 concentration in each sample is interpolated from this standard curve.

 NCBI Summary for MS4A1    This gene encodes a member of the membrane-spanning 4A gene family. Members of this nascent protein family are characterized by common structural features and similar intron/exon splice boundaries and display unique expression patterns among hematopoietic cells and nonlymphoid tissues. This gene encodes a B-lymphocyte surface molecule which plays a role in the development and differentiation of B-cells into plasma cells. This family member is localized to 11q12, among a cluster of family members. Alternative splicing of this gene results in two transcript variants which encode the same protein. Defects in MS4A1 are the cause of immunodeficiency common variable type 5 (CVID5); also called antibody deficiency due to CD20 defect. CVID5 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B-cells is usually in the normal range, but can be low.
