Intended Uses    This Quantitative Sandwich ELISA kit is only for in vitro research use only, not for drug, household, therapeutic or diagnostic applications! It is intended to be determinated GP6 concentrations in Canine serum, plasma and other body fluids. Using Purified Canine GP6 antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add GP6 and GP6 antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of GP6 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
 Intra-assay Precision    Intra-assay CV (%) is less than 15%.
 Inter-assay Precision    Inter-assay CV (%) is less than 15%. [CV(%) = SD/mean x100]

Preparation and Storage    Store all reagents at 2-8 degree C
Other Notes    Small volumes of GP6 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for GP6 purchase    MBS005646 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Glycoprotein VI, Platelet (GP6) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 6.25 ng/ml - 200 ng/ml in biological research samples containing GP6, with an estimated sensitivity of 1.0 ng/ml. The ELISA analytical biochemical technique of the MBS005646 kit is based on GP6 antibody-GP6 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GP6 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GP6. Appropriate sample types may include undiluted human body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Tissue Homogenate, Feces, Urine and Body Fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

NCBI Summary for GP6    This gene encodes a platelet membrane glycoprotein of the immunoglobulin superfamily. The encoded protein is a receptor for collagen and plays a critical role in collagen-induced platelet aggregation and thrombus formation. The encoded protein forms a complex with the Fc receptor gamma-chain that initiates the platelet activation signaling cascade upon collagen binding. Mutations in this gene are a cause of platelet-type bleeding disorder-11 (BDPLT11). Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Dec 2011]
Section Bar
UniProt Comments for GP6    GPVI: Collagen receptor involved in collagen-induced platelet adhesion and activation. Plays a key role in platelet procoagulant activity and subsequent thrombin and fibrin formation. This procoagulant function may contribute to arterial and venous thrombus formation. The signaling pathway involves the FcR gamma- chain, the Src kinases (likely Fyn/Lyn), the adapter protein LAT and leads to the activation of phospholipase C gamma2. Defects in GP6 are the cause of bleeding disorder platelet-type 11 (BDPLT11). BDPLT11 is a mild to moderate bleeding disorder caused by defective platelet activation and aggregation in response to collagen. 3 isoforms of the human protein are produced by alternative splicing.
