The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.5 ng/ml ~ 16 ng/ml in biological research samples containing TPS. The ELISA analytical biochemical technique of the MBS013118 kit is based on TPS antibody-TPS antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TPS antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TPS. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Cell Culture Supernatants, Tissue Homogenate, Urine and other Body Fluids. Product Description specifically for TPS elisa kit

   Principle of the Assay: This ELISA kit is intended for laboratory in vitro research use only, not for drug, household or other use! The Stop Solution changes the color from blue to yellow and the intensity of the color is measured at 450 nm by a spectrophotometer. In order to measure the concentration of Canine TPS in the specimen, this ELISA kit includes a set of calibration standards. The calibration standards are assayed at the same time as the specimens and allow the operator to produce a standard curve of Optical Density versus Canine TPS concentration. The concentration of Canine TPS in the specimens is then determined by comparing the O.D. of the specimens to the standard curve.

Background: ELISA is a simple and highly sensitive method of analysis that allows for simultaneous and rapid quantification of a large number of specimens. The assay is based on the specific recognition of the target compound (analyte/antigen) by antibodies which bind to the compound. The antigen-antibody complex is detected and measured with the aid of an enzyme-labeled antibody or antigen. Upon addition of a non-colored reagent, the enzyme produces a color reaction where the color intensity is directly or inversely proportional to the concentration of the analyte in the specimen.

 NCBI Summary for TPS    Tryptases comprise a family of trypsin-like serine proteases, the peptidase family S1. Tryptases are enzymatically active only as heparin-stabilized tetramers, and they are resistant to all known endogenous proteinase inhibitors. Several tryptase genes are clustered on chromosome 16p13.3. These genes are characterized by several distinct features. They have a highly conserved 3' UTR and contain tandem repeat sequences at the 5' flank and 3' UTR which are thought to play a role in regulation of the mRNA stability. These genes have an intron immediately upstream of the initiator Met codon, which separates the site of transcription initiation from protein coding sequence. This feature is characteristic of tryptases but is unusual in other genes. The alleles of this gene exhibit an unusual amount of sequence variation, such that the alleles were once thought to represent two separate genes, alpha and beta 1. Beta tryptases appear to be the main isoenzymes expressed in mast cells; whereas in basophils, alpha tryptases predominate.
