Specificity    This assay has high sensitivity and excellent specificity for detection of ZP3. No significant cross-reactivity or interference between ZP3 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ZP3 and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of ZP3 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for ZP3 purchase    MBS7202994 is a ready-to-use microwell, strip plate Quantitative Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Zona pellucida sperm-binding protein 3 (ZP3) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ZP3, with an estimated sensitivity of 1.0 ng/mL. The ELISA analytical biochemical technique of the MBS7202994 kit is based on ZP3 antibody-ZP3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ZP3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ZP3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for ZP3 elisa kit

Intended Uses: This ZP3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine ZP3. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: ZP3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ZP3 antibody and an ZP3-HRP conjugate. The assay sample and buffer are incubated together with ZP3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ZP3 concentration since ZP3 from samples and ZP3-HRP conjugate compete for the anti-ZP3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ZP3 from the sample, fewer sites are left to bind ZP3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ZP3 concentration in each sample is interpolated from this standard curve.

NCBI Summary for ZP3    The zona pellucida is an extracellular matrix that surrounds the oocyte and early embryo. It is composed primarily of three or four glycoproteins with various functions during fertilization and preimplantation development. The protein encoded by this gene is a structural component of the zona pellucida and functions in primary binding and induction of the sperm acrosome reaction. The nascent protein contains a N-terminal signal peptide sequence, a conserved ZP domain, a C-terminal consensus furin cleavage site, and a transmembrane domain. It is hypothesized that furin cleavage results in release of the mature protein from the plasma membrane for subsequent incorporation into the zona pellucida matrix. However, the requirement for furin cleavage in this process remains controversial based on mouse studies. A variation in the last exon of this gene has previously served as the basis for an additional ZP3 locus; however, sequence and literature review reveals that there is only one full-length ZP3 locus in the human genome. Another locus encoding a bipartite transcript designated POMZP3 contains a duplication of the last four exons of ZP3, including the above described variation, and maps closely to this gene.  UniProt Comments for ZP3    ZP3: The mammalian zona pellucida, which mediates species- specific sperm binding, induction of the acrosome reaction and prevents post-fertilization polyspermy, is composed of three to four glycoproteins, ZP1, ZP2, ZP3, and ZP4. ZP3 is essential for sperm binding and zona matrix formation. Belongs to the ZP domain family. ZPC subfamily. 3 isoforms of the human protein are produced by alternative splicing.
