Specificity    This assay has high sensitivity and excellent specificity for detection of Chicken EPP. No significant cross-reactivity or interference between Chicken EPP and analogues was observed. Note: Limited by current skills and knowledge, it is impossible for us to complete the cross- reactivity detection between Chicken EPP and all the analogues, therefore, cross reaction may still exist.

Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay.
Other Notes    Small volumes of EPP elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for EPP purchase    MBS2512333 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the EPP, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing EPP. The ELISA analytical biochemical technique of the MBS2512333 kit is based on EPP antibody-EPP antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect EPP antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, EPP. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Biological Fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for EPP elisa kit

Description: The kit is a competitive enzyme immunoassay for in vitro quantitative measurement of EPP in Chicken serum, plasma and other biological fluids
Principle of the Assay: This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with Chicken EPP. During the reaction, Chicken EPP in the sample or standard competes with a fixed amount of Chicken EPP on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Chicken EPP. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Chicken EPP in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Summary for EPP    This gene is a member of the peroxidase gene family and is expressed in eosinophils. The encoded precursor protein is processed into covalently attached heavy and light chains to form the mature enzyme, which functions as an oxidant. The enzyme is released at sites of parasitic infection or allergen stimulation to mediate lysis of protozoa or parasitic worms. The gene is found in a cluster of three peroxidase genes at chromosome 17q23. Mutations in this gene result in eosinophil peroxidase deficiency. [provided by RefSeq, Sep 2009]
Section Bar
UniProt Comments for EPP    EPX: Mediates tyrosine nitration of secondary granule proteins in mature resting eosinophils. Shows significant inhibitory activity towards Mycobacterium tuberculosis H37Rv by inducing bacterial fragmentation and lysis. Defects in EPX are the cause of eosinophil peroxidase deficiency (EPD). EPD is an autosomal recessive defect where anomalous eosinophils are characterized by nuclear hypersegmentation, hypogranulation, and negative peroxidase and phospholipid staining. Belongs to the peroxidase family. XPO subfamily.

Protein type: Oxidoreductase; EC 1.11.1.7

Chromosomal Location of Human Ortholog: 17q23.1

Molecular Function: peroxidase activity; metal ion binding; heme binding

Biological Process: positive regulation of interleukin-4 production; negative regulation of interleukin-10 production; hydrogen peroxide catabolic process; negative regulation of interleukin-5 production; response to oxidative stress; defense response to nematode

Disease: Eosinophil Peroxidase Deficiency
