It is intended to be determinated PNFKB concentrations in Fish serum, plasma and other body fluids. Using Purified Fish PNFKB antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add PNFKB and PNFKB antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of PNFKB in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 62.5 pg/ml - 2000 pg/ml in biological research samples containing PNFKB, with an estimated sensitivity of 10 pg/ml. Kit is applicable for testing variable types of samples, including plasma, serum and other bodily fluids, and also tissue homogenates.

Product contains microwell plate pre-coated with purified capture antibodies against PNFKB and all reagents required for its quantification, including sample diluent and wash solution.

Assay kits provide colorimetric detection of PNFKB using detector antibodies against PNFKB labeled with HRP as signaling molecule, and TMB as its substrate solution.

Minimal detectable concentration of ELISA kit is 10pg/mL and its linear range is 62.5-2,000pg/mL; calibrators for standard curve include six levels.

This protein is an inhibitor of smooth muscle myosin phosphatase, and has higher inhibitory activity when phosphorylated.  UniProt Comments for PNFKB    PPP1R14A: a phosphorylation-dependent inhibitory subunit of protein phosphatase 1 (PP-1A). Phosphorylation by PKC regulates its binding to PP-1A. Acts co-operatively to inhibit myosin light chain phosphatase (MLCP) activity. Inhibition leads to increased myosin phosphorylation and enhances smooth muscle contraction in the absence of increased intracellular Ca(2 ) concentration. Involved in the regulation of cell morphology, re-organisation of microfilaments, and cell spreading. Also expressed in human platelets and in brain where it may play a role in cerebellar long-term synaptic depression. Phosphorylation by ILK or Rho kinase enhances its ability to inhibit PP-1A.
