Preparation and Storage

Intended Uses: The kit is a competitive inhibition enzyme immunoassay technique for the in vitro quantitative measurement of DHVD3 in general serum, plasma or other biological fluids.
Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to DHVD3 has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled DHVD3 and unlabeled DHVD3 (Standards or samples) with the pre-coated antibody specific to DHVD3. After incubation the unbound conjugate is washed off. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of DHVD3 in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of DHVD3 in the sample.

This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This mitochondrial protein initiates the degradation of 1,25-dihydroxyvitamin D3, the physiologically active form of vitamin D3, by hydroxylation of the side chain. In regulating the level of vitamin D3, this enzyme plays a role in calcium homeostasis and the vitamin D endocrine system. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
