Specificity    This assay has high sensitivity and excellent specificity for detection of PLOD1. No significant cross-reactivity or interference between PLOD1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between PLOD1 and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of PLOD1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PLOD1 purchase    MBS7223151 is a ready-to-use microwell, strip plate Quantitative Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Procollagen-lysine, 2-oxoglutarate 5-dioxygenase 1 (PLOD1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PLOD1, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS7223151 kit is based on PLOD1 antibody-PLOD1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PLOD1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PLOD1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for PLOD1 elisa kit

Intended Uses: This PLOD1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Guinea pig PLOD1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: PLOD1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-PLOD1 antibody and an PLOD1-HRP conjugate. The assay sample and buffer are incubated together with PLOD1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PLOD1 concentration since PLOD1 from samples and PLOD1-HRP conjugate compete for the anti-PLOD1 antibody binding site. Since the number of sites is limited, as more sites are occupied by PLOD1 from the sample, fewer sites are left to bind PLOD1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PLOD1 concentration in each sample is interpolated from this standard curve.

NCBI Summary for PLOD1    Lysyl hydroxylase is a membrane-bound homodimeric protein localized to the cisternae of the endoplasmic reticulum. The enzyme (cofactors iron and ascorbate) catalyzes the hydroxylation of lysyl residues in collagen-like peptides. The resultant hydroxylysyl groups are attachment sites for carbohydrates in collagen and thus are critical for the stability of intermolecular crosslinks. Some patients with Ehlers-Danlos syndrome type VI have deficiencies in lysyl hydroxylase activity. [provided by RefSeq, Jul 2008]
Section Bar
UniProt Comments for PLOD1    PLOD1: Forms hydroxylysine residues in -Xaa-Lys-Gly- sequences in collagens. These hydroxylysines serve as sites of attachment for carbohydrate units and are essential for the stability of the intermolecular collagen cross-links. Defects in PLOD1 are the cause of Ehlers-Danlos syndrome type 6 (EDS6). EDS is a connective tissue disorder characterized by hyperextensible skin, atrophic cutaneous scars due to tissue fragility and joint hyperlaxity. EDS6 is characterized by the presence of ocular complications, particularly retinal detachment. Defects in PLOD1 are the cause of Nevo syndrome (NEVOS). This is a rare, autosomal recessive disorder characterized by increased perinatal length, kyphosis, muscular hypotonia, and joint laxity. Nevo syndrome and EDS-VI have similar clinical phenotypes. Some authors consider that both syndromes are the same clinical entity.
