Specificity    This assay has high sensitivity and excellent specificity for detection of PTPRR. No significant cross-reactivity or interference between PTPRR and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between PTPRR and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay.
Other Notes    Small volumes of PTPRR elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PTPRR purchase    MBS720125 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Protein Tyrosine Phosphatase Receptor Type R (PTPRR) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PTPRR, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS720125 kit is based on PTPRR antibody-PTPRR antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PTPRR antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PTPRR. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for PTPRR elisa kit

Principle of the assay: PTPRR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PTPRR antibody and an PTPRR-HRP conjugate. The assay sample and buffer are incubated together with PTPRR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PTPRR concentration since PTPRR from samples and PTPRR-HRP conjugate compete for the anti-PTPRR antibody binding site. Since the number of sites is limited, as more sites are occupied by PTPRR from the sample, fewer sites are left to bind PTPRR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PTPRR concentration in each sample is interpolated from this standard curve.

NCBI Summary for PTPRR    The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP possesses an extracellular region, a single transmembrane region, and a single intracellular catalytic domain, and thus represents a receptor-type PTP. Silencing of this gene has been associated with colorectal cancer. Multiple transcript variants encoding different isoforms have been found for this gene. This gene shares a symbol (PTPRQ) with another gene, protein tyrosine phosphatase, receptor type, Q (GeneID 374462), which is also located on chromosome 12. [provided by RefSeq, May 2011]

UniProt Comments for PTPRR    PTP-SL: Sequesters mitogen-activated protein kinases (MAPKs) such as MAPK1, MAPK3 and MAPK14 in the cytoplasm in an inactive form. The MAPKs bind to a dephosphorylated kinase interacting motif, phosphorylation of which by the protein kinase A complex releases the MAPKs for activation and translocation into the nucleus. Belongs to the protein-tyrosine phosphatase family. Receptor class 7 subfamily. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 3.1.3.48; Receptor protein phosphatase, tyrosine; Motility/polarity/chemotaxis; Membrane protein, integral

Chromosomal Location of Human Ortholog: 12q15

Cellular Component: extracellular space; perinuclear region of cytoplasm; plasma membrane; integral to membrane

Molecular Function: protein binding; transmembrane receptor protein tyrosine phosphatase activity; protein tyrosine phosphatase activity; protein kinase binding

Biological Process: in utero embryonic development; protein amino acid dephosphorylation
