Product Description specifically for CD209 elisa kit

   Intended Uses: This CD209 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CD209. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: CD209 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CD209 antibody and an CD209-HRP conjugate. The assay sample and buffer are incubated together with CD209-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD209 concentration since CD209 from samples and CD209-HRP conjugate compete for the anti-CD209 antibody binding site. Since the number of sites is limited, as more sites are occupied by CD209 from the sample, fewer sites are left to bind CD209-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD209 concentration in each sample is interpolated from this standard curve.

 NCBI Summary for CD209    This gene encodes a transmembrane receptor and is often referred to as DC-SIGN because of its expression on the surface of dendritic cells and macrophages. The encoded protein is involved in the innate immune system and recognizes numerous evolutionarily divergent pathogens ranging from parasites to viruses with a large impact on public health. The protein is organized into three distinct domains: an N-terminal transmembrane domain, a tandem-repeat neck domain and C-type lectin carbohydrate recognition domain. The extracellular region consisting of the C-type lectin and neck domains has a dual function as a pathogen recognition receptor and a cell adhesion receptor by binding carbohydrate ligands on the surface of microbes and endogenous cells. The neck region is important for homo-oligomerization which allows the receptor to bind multivalent ligands with high avidity. Variations in the number of 23 amino acid repeats in the neck domain of this protein are rare but have a significant impact on ligand binding ability. This gene is closely related in terms of both sequence and function to a neighboring gene (GeneID 10332; often referred to as L-SIGN). DC-SIGN and L-SIGN differ in their ligand-binding properties and distribution. Alternative splicing results in multiple variants.[provided by RefSeq, Feb 2009]

Thought to mediate the endocytosis of pathogens which are subsequently degraded in lysosomal compartments.
