The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 31.2 pg/ml-2000 pg/ml in biological research samples containing CT-1, with an estimated sensitivity of < 10 pg/ml. The ELISA analytical biochemical technique of the MBS355361 kit is based on CT-1 antibody-CT-1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CT-1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CT-1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

   Principle of the assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-CT-1 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-CT-1 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the CT-1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of CT-1 can be calculated.

Background: Cardiotrophin-1 (CT-1) is a cytokine. CTF1 gene contains 3 exons, spans over 6 to 7 kb. It is a cardiac hypertrophic factor of 21.5 kDa and a protein member of the IL-6 cytokine family. It is highly expressed in the heart, skeletal muscle, prostate and ovary and to lower levels in lung, kidney, pancreas, thymus, testis and small intestine. CT-1 is associated with the pathophysiology of heart diseases, including hypertension, myocardial infarction, valvular heart disease, and congestive heart failure. This protein exerts its cellular effects by interacting with the glycoprotein 130 (gp130)/leukemia inhibitory factor receptor beta (LIFR) heterodimer.
