   Intended Uses: This CASP14 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CASP14. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay||CASP14 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CASP14 antibody and an CASP14-HRP conjugate. The assay sample and buffer are incubated together with CASP14-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CASP14 concentration since CASP14 from samples and CASP14-HRP conjugate compete for the anti-CASP14 antibody binding site. Since the number of sites is limited, as more sites are occupied by CASP14 from the sample, fewer sites are left to bind CASP14-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CASP14 concentration in each sample is interpolated from this standard curve.

 NCBI Summary for CASP14    This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This caspase has been shown to be processed and activated by caspase 8 and caspase 10 in vitro, and by anti-Fas agonist antibody or TNF-related apoptosis inducing ligand in vivo. The expression and processing of this caspase may be involved in keratinocyte terminal differentiation, which is important for the formation of the skin barrier. [provided by RefSeq, Jul 2008]
