The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 78.125-5000pg/mL in biological research samples containing CHIT1, with an estimated sensitivity of Min: 46.875pg/mL; Max: 5000pg/mL. The ELISA analytical biochemical technique of the MBS2533435 kit is based on CHIT1 antibody-CHIT1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CHIT1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CHIT1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Biological Fluids.    Intended Uses: This ELISA kit applies to the invitro quantitative determination of HumanCHIT1concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to CHIT1. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for CHIT1and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain CHIT1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of CHIT1.You can calculate the concentration of CHIT1in the samples by comparing the OD of the samples to the standard curve.

 NCBI Summary for CHIT1    Chitotriosidase is secreted by activated human macrophages and is markedly elevated in plasma of Gaucher disease patients. The expression of chitotriosidase occurs only at a late stage of differentiation of monocytes to activated macrophages in culture. Human macrophages can synthesize a functional chitotriosidase, a highly conserved enzyme with a strongly regulated expression. This enzyme may play a role in the degradation of chitin-containing pathogens. Several alternatively spliced transcript variants have been described for this gene. [provided by RefSeq, Jan 2012]
