The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.5-10ng/mL in biological research samples containing B7 H4, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS751659 kit is based on B7 H4 antibody-B7 H4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect B7 H4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, B7 H4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

   Priciple of the assay: B7H4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-B7H4 antibody and an B7H4-HRP conjugate. The assay sample and buffer are incubated together with B7H4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the B7H4 concentration since B7H4 from samples and B7H4-HRP conjugate compete for the anti-B7H4 antibody binding site. Since the number of sites is limited, as more sites are occupied by B7H4 from the sample, fewer sites are left to bind B7H4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The B7H4 concentration in each sample is interpolated from this standard curve.

 NCBI Summary for B7 H4    This gene encodes a protein belonging to the B7 costimulatory protein family. Proteins in this family are present on the surface of antigen-presenting cells and interact with ligand bound to receptors on the surface of T cells. Studies have shown that high levels of the encoded protein has been correlated with tumor progression. A pseudogene of this gene is located on chromosome 20.
