Preparation and Storage    For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C.

For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal.

The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay.
Other Notes    Small volumes of CUBN elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for CUBN purchase    MBS2021105 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Cubilin (CUBN) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.781-50ng/mL in biological research samples containing CUBN, with an estimated sensitivity of Typically less than 0.34ng/mL. The ELISA analytical biochemical technique of the MBS2021105 kit is based on CUBN antibody-CUBN antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CUBN antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CUBN. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Tissue homogenates and other biological fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for CUBN elisa kit

Principle of the Assay: The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cubilin (CUBN). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Cubilin (CUBN). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Cubilin (CUBN), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Cubilin (CUBN) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

UniProt Comments for CUBN    CUBN: Cotransporter which plays a role in lipoprotein, vitamin and iron metabolism, by facilitating their uptake. Binds to ALB, MB, Kappa and lambda-light chains, TF, hemoglobin, GC, SCGB1A1, APOA1, high density lipoprotein, and the GIF-cobalamin complex. The binding of all ligands requires calcium. Serves as important transporter in several absorptive epithelia, including intestine, renal proximal tubules and embryonic yolk sac. Interaction with LRP2 mediates its trafficking throughout vesicles and facilitates the uptake of specific ligands like GC, hemoglobin, ALB, TF and SCGB1A1. Interaction with AMN controls its trafficking to the plasma membrane and facilitates endocytosis of ligands. May play an important role in the development of the peri-implantation embryo through internalization of APOA1 and cholesterol. Binds to LGALS3 at the maternal-fetal interface. Defects in CUBN are a cause of recessive hereditary megaloblastic anemia 1 (RH-MGA1); also known as MGA1 Norwegian type or Imerslund-Grasbeck syndrome (I-GS). RH-MGA1 is due to selective malabsorption of vitamin B12. Defects in vitamin B12 absorption lead to impaired function of thymidine synthase. As a consequence DNA synthesis is interrupted. Rapidly dividing cells involved in erythropoiesis are particularly affected.

Cellular Component: Golgi apparatus; lysosomal lumen; protein complex; lysosomal membrane; endoplasmic reticulum; brush border membrane; coated pit; membrane; apical part of cell; endocytic vesicle; cytoplasm; apical plasma membrane; endosome membrane; brush border; endosome

Molecular Function: identical protein binding; protein homodimerization activity; hemoglobin binding; cobalamin transporter activity; calcium ion binding; receptor activity; cobalamin binding

Biological Process: receptor-mediated endocytosis; cholesterol metabolic process; vitamin metabolic process; in utero embryonic development; lipoprotein transport; cobalamin transport; hemoglobin import; response to nutrient

Research Articles on CUBN    1. We conclude that the megalin/cubilin/amnionless/Dab2 complex/es participate in intestinal processes, mainly during the lactation period and that Dab2 may act as an adaptor in intestinal processes mediated by ApoER2 and VLDLR.
