The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.313-20ng/mL in biological research samples containing DT, with an estimated sensitivity of Min: 0.188ng/mL; Max: 20ng/mL. The ELISA analytical biochemical technique of the MBS2509575 kit is based on DT antibody-DT antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect DT antigen targets in samples.    Description: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of DT in human serum, plasma and other biological fluids
Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human DT. Standards or samples are added to the appropriate micro ELISA plate wells and bound by the specific antibody. Then a biotinylated detection antibody specific for Human DT and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human DT, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of Human DT. You can calculate the concentration of Human DT in the samples by comparing the OD of the samples to the standard curve.

 NCBI Summary for DT    This gene encodes a member of the plakin protein family of adhesion junction plaque proteins. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for this gene, but the full-length nature of some variants has not been defined. It has been reported that some isoforms are expressed in neural and muscle tissue, anchoring neural intermediate filaments to the actin cytoskeleton, and some isoforms are expressed in epithelial tissue, anchoring keratin-containing intermediate filaments to hemidesmosomes. Consistent with the expression, mice defective for this gene show skin blistering and neurodegeneration. [provided by RefSeq, Mar 2010]
