The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 3.125-200pg/mL in biological research samples containing DEFa5, with an estimated sensitivity of 1.31pg/mL. The ELISA analytical biochemical technique of the MBS455303 kit is based on DEFa5 antibody-DEFa5 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect DEFa5 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, DEFa5. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of DEFa5 in human serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to DEFa5. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to DEFa5. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain DEFa5, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of DEFa5 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

|
Defensins are a family of antimicrobial and cytotoxic peptides thought to be involved in host defense. They are abundant in the granules of neutrophils and also found in the epithelia of mucosal surfaces such as those of the intestine, respiratory tract, urinary tract, and vagina. Members of the defensin family are highly similar in protein sequence and distinguished by a conserved cysteine motif. Several of the alpha defensin genes appear to be clustered on chromosome 8. The protein encoded by this gene, defensin, alpha 5, is highly expressed in the secretory granules of Paneth cells of the ileum. [provided by RefSeq, Oct 2014]
