Specificity    This assay has high sensitivity and excellent specificity for detection of human EWSR1. No significant cross-reactivity or interference between human EWSR1 and analogues was observed.
Section Bar
Samples    Serum, plasma, tissue homogenates, Cell lysates
 Assay Type    Sandwich
 Detection Range    25 pg/ml -1600 pg/ml.
Sensitivity    The minimum detectable dose of human EWSR1 is typically less than 6.25 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision    Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision    Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Preparation and Storage    Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
ISO Certification    Manufactured in an ISO 9001:2008 Certified Laboratory.
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of EWSR1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for EWSR1 purchase    MBS908129 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Ewing sarcoma breakpoint region 1 (EWSR1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 25 pg/ml -1600 pg/ml in biological research samples containing EWSR1, with an estimated sensitivity of The minimum detectable dose of human EWSR1 is typically less than 6.25 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. The ELISA analytical biochemical technique of the MBS908129 kit is based on EWSR1 antibody-EWSR1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect EWSR1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, EWSR1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates, Cell lysates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for EWSR1 elisa kit

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for EWSR1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EWSR1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EWSR1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EWSR1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Summary for EWSR1    This gene encodes a multifunctional protein that is involved in various cellular processes, including gene expression, cell signaling, and RNA processing and transport. The protein includes an N-terminal transcriptional activation domain and a C-terminal RNA-binding domain. Chromosomal translocations between this gene and various genes encoding transcription factors result in the production of chimeric proteins that are involved in tumorigenesis. These chimeric proteins usually consist of the N-terminal transcriptional activation domain of this protein fused to the C-terminal DNA-binding domain of the transcription factor protein. Mutations in this gene, specifically a t(11;22)(q24;q12) translocation, are known to cause Ewing sarcoma as well as neuroectodermal and various other tumors. Alternative splicing of this gene results in multiple transcript variants. Related pseudogenes have been identified on chromosomes 1 and 14. [provided by RefSeq, Jul 2009]

UniProt Comments for EWSR1    EWS: an oncogenic RNA-binding protein. Might normally function as a repressor. Binds RPB3, SF1, calmodulin and RNA. Interacts with and regulated by Pyk2. Relocates from cytoplasm to ribosomes upon Pyk2 activation. EWS-fusion-proteins (EFPS) may play a role in the tumorigenic process. They may disturb gene expression by mimicking, or interfering with the normal function of CTD-POLII within the transcription initiation complex. 5 alternatively-spliced isoforms have been described.

Protein type: Transcription, coactivator/corepressor; Oncoprotein; RNA-binding

Chromosomal Location of Human Ortholog: 22q12.2
