   Intended Uses: For the quantitative detection of Human Hypoxia-inducible factor 2 (HIF-2) concentration in serum, plasma, cell lysates and other biological fluids.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate HIF-2 in Human serum, plasma, cell lysates. An antibody specific for HIF-2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HIF-2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HIF-2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HIF-2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

 NCBI Summary for HIF-2    This gene encodes a transcription factor involved in the induction of genes regulated by oxygen, which is induced as oxygen levels fall. The encoded protein contains a basic-helix-loop-helix domain protein dimerization domain as well as a domain found in proteins in signal transduction pathways which respond to oxygen levels. Binds to core DNA sequence 5'-[AG]CGTG-3' within the hypoxia response element (HRE) of target gene promoters.  Research Articles on HIF-2    1. Knockdown of either HIF-1 or CREB or both in hypoxia reduced the expression of hypoxia-response elements- and CRE-mediated gene expression, diminished cell proliferation and increased caspase-3 activity. We did not detect any significant effect of the efficiently knocked down HIF-2 on any of the functions tested in vitro.
