The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 23.5 pg/ml-1500 pg/ml in biological research samples containing IMPDH2, with an estimated sensitivity of 5.8 pg/ml. The ELISA analytical biochemical technique of the MBS906235 kit is based on IMPDH2 antibody-IMPDH2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect IMPDH2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, IMPDH2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernates, tissue homogenates. Product Description specifically for IMPDH2 elisa kit

   Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to IMPDH2. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for IMPDH2 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain IMPDH2, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of IMPDH2 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

 NCBI Summary for IMPDH2    This gene encodes the rate-limiting enzyme in the de novo guanine nucleotide biosynthesis. It is thus involved in maintaining cellular guanine deoxy- and ribonucleotide pools needed for DNA and RNA synthesis. The encoded protein catalyzes the NAD-dependent oxidation of inosine-5'-monophosphate into xanthine-5'-monophosphate, which is then converted into guanosine-5'-monophosphate. This gene is up-regulated in some neoplasms, suggesting it may play a role in malignant transformation. [provided by RefSeq, Jul 2008]
