Preparation and Storage    For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C.

For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal.

The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of LIPE elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for LIPE purchase    MBS2021827 is a ready-to-use microwell, strip plate Quantitative Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Lipase, Hormone Sensitive (LIPE) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.156-10ng/mL in biological research samples containing LIPE, with an estimated sensitivity of <0.065ng/mL. The ELISA analytical biochemical technique of the MBS2021827 kit is based on LIPE antibody-LIPE antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LIPE antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LIPE. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Human serum, plasma and other biological fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for LIPE elisa kit

Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of LIPE in human serum, plasma and other biological fluids.

Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to LIPE. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to LIPE. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain LIPE, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of LIPE in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Summary for LIPE    The protein encoded by this gene has a long and a short form, generated by use of alternative translational start codons. The long form is expressed in steroidogenic tissues such as testis, where it converts cholesteryl esters to free cholesterol for steroid hormone production. The short form is expressed in adipose tissue, among others, where it hydrolyzes stored triglycerides to free fatty acids. [provided by RefSeq, Jul 2008]

UniProt Comments for LIPE    HSL: hormone sensitive lipase is a lipolytic enzyme of the 'GDXG' family. Plays a rate limiting step in triglyceride lipolysis. In adipose tissue and heart, it primarily hydrolyzes stored triglycerides to free fatty acids, while in steroidogenic tissues, it principally converts cholesteryl esters to free cholesterol for steroid hormone production. Rapidly activated by cAMP-dependent phosphorylation under the influence of catecholamines. Dephosphorylation and inactivation are controlled by insulin.
