Searchable Terms for MIF purchase    MBS355376 is a ready-to-use microwell, strip-or-full plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the MIF, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 156 pg/ml-10,000 pg/ml in biological research samples containing MIF, with an estimated sensitivity of < 20pg/ml. The ELISA analytical biochemical technique of the MBS355376 kit is based on MIF antibody-MIF antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MIF antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MIF. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for MIF elisa kit

Background/Introduction: Macrophage migration inhibitory factor (MIF or MMIF) also known as glycosylation-inhibiting factor (GIF), L-dopachrome isomerase, or phenylpyruvate tautomerase is a protein that in humans is encoded by the MIF gene. Macrophage migration inhibitory factor assembles into a trimer composed of three identical subunits. Each of these monomers contain two antiparallel alpha helices and a four-stranded beta sheet. The monomers surround a central channel with 3-fold rotational symmetry. MIF plays a role in the regulation of macrophage function in host defense through the suppression of anti-inflammatory effects of glucocorticoids. It is an inflammatory mediator associated with rheumatoid arthritis (RA) severity. Additionally, evidence suggests that there is a correlation between MIF production and metastatic potential in colorectal cancer.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-MIF polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-MIF polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the MIF amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of MIF can be calculated.

NCBI Summary for MIF    This gene encodes a lymphokine involved in cell-mediated immunity, immunoregulation, and inflammation. It plays a role in the regulation of macrophage function in host defense through the suppression of anti-inflammatory effects of glucocorticoids. This lymphokine and the JAB1 protein form a complex in the cytosol near the peripheral plasma membrane, which may indicate an additional role in integrin signaling pathways.  UniProt Comments for MIF    Function: Pro-inflammatory cytokine. Involved in the innate immune response to bacterial pathogens. The expression of MIF at sites of inflammation suggests a role as mediator in regulating the function of macrophages in host defense. Counteracts the anti-inflammatory activity of glucocorticoids. Has phenylpyruvate tautomerase and dopachrome tautomerase activity (in vitro), but the physiological substrate is not known. It is not clear whether the tautomerase activity has any physiological relevance, and whether it is important for cytokine activity. Ref.23 Ref.24

Catalytic activity: Keto-phenylpyruvate = enol-phenylpyruvate. Ref.32 Ref.34L-dopachrome = 5,6-dihydroxyindole-2-carboxylate. Ref.32 Ref.34

Subunit structure: Homotrimer. Interacts with CXCR2 extracellular domain

By similarity. Interacts with the CD74 extracellular domain, COPS5 and BNIPL. Ref.18 Ref.21 Ref.22 Ref.25 Ref.33 Ref.34

Subcellular location: Secreted. Cytoplasm. Note: Does not have a cleavable signal sequence and is secreted via a specialized, non-classical pathway. Secreted by macrophages upon stimulation by bacterial lipopolysaccharide (LPS), or by M.tuberculosis antigens. Ref.1 Ref.2 Ref.18 Ref.23 Ref.25

Induction: Up-regulated in concanavalin-A-treated lymphocytes. Up-regulated in macrophages upon exposure to M.tuberculosis antigens. Ref.1 Ref.23

Involvement in disease: Rheumatoid arthritis systemic juvenile (RASJ) [MIM:604302]: An inflammatory articular disorder with systemic-onset beginning before the age of 16. It represents a subgroup of juvenile arthritis associated with severe extraarticular features and occasionally fatal complications. During active phases of the disorder, patients display a typical daily spiking fever, an evanescent macular rash, lymphadenopathy, hepatosplenomegaly, serositis, myalgia and arthritis.Note: Disease susceptibility is associated with variations affecting the gene represented in this entry.

Miscellaneous: Serum levels of MIF are elevated in patients with severe sepsis or septic shock. High levels of MIF are correlated with low survival. Drugs that inhibit tautomerase activity protect against death due to sepsis.

Sequence similarities: Belongs to the MIF family.

Research Articles on MIF    1. Mutant N110C can form a higher-order oligomer in equilibrium with a single locked trimer. The X-ray structure confirms a local conformational change that disrupts the subunit interface and results in global changes responsible for the oligomeric form.
