Specificity    This assay has high sensitivity and excellent specificity for detection of MTNR-1A. No significant cross-reactivity or interference between MTNR-1A and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between MTNR-1A and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of MTNR1A elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for MTNR1A purchase    MBS7229402 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Melatonin receptor type 1A (MTNR1A) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.5-10ng/mL in biological research samples containing MTNR1A, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS7229402 kit is based on MTNR1A antibody-MTNR1A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MTNR1A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MTNR1A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for MTNR1A elisa kit

Intended Uses: This MTNR-1A ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human MTNR-1A. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay||MTNR-1A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MTNR-1A antibody and an MTNR-1A-HRP conjugate. The assay sample and buffer are incubated together with MTNR-1A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MTNR-1A concentration since MTNR-1A from samples and MTNR-1A-HRP conjugate compete for the anti-MTNR-1A antibody binding site. Since the number of sites is limited, as more sites are occupied by MTNR-1A from the sample, fewer sites are left to bind MTNR-1A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MTNR-1A concentration in each sample is interpolated from this standard curve.

NCBI Summary for MTNR1A    This gene encodes one of two high affinity forms of a receptor for melatonin, the primary hormone secreted by the pineal gland. This receptor is a G-protein coupled, 7-transmembrane receptor that is responsible for melatonin effects on mammalian circadian rhythm and reproductive alterations affected by day length. The receptor is an integral membrane protein that is readily detectable and localized to two specific regions of the brain. The hypothalamic suprachiasmatic nucleus appears to be involved in circadian rhythm while the hypophysial pars tuberalis may be responsible for the reproductive effects of melatonin. [provided by RefSeq, Jul 2008]

Research Articles on MTNR1A    1. Suggest that physiological regulation of the melatonin receptors may result from complex and subtle mechanisms, a small difference in affinity between the active and inactive states of the receptor, and spontaneous coupling to G-proteins.
