The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 250-5000pg/mL in biological research samples containing MLR Bf, with an estimated sensitivity of 1.0pg/mL. The ELISA analytical biochemical technique of the MBS756357 kit is based on MLR Bf antibody-MLR Bf antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MLR Bf antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MLR Bf. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: MLR-Bf ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MLR-Bf antibody and an MLR-Bf-HRP conjugate. The assay sample and buffer are incubated together with MLR-Bf-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MLR-Bf concentration since MLR-Bf from samples and MLR-Bf-HRP conjugate compete for the anti-MLR-Bf antibody binding site. Since the number of sites is limited, as more sites are occupied by MLR-Bf from the sample, fewer sites are left to bind MLR-Bf-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MLR-Bf concentration in each sample is interpolated from this standard curve.
