Intended Uses    This Quantitative Sandwich ELISA kit is only for in vitro research use only, not for drug, household, therapeutic or diagnostic applications! It is intended to be determinated NADHPOX concentrations in Human serum, plasma and other body fluids. Using Purified Human NADHPOX antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add NADHPOX and NADHPOX antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of NADHPOX in the samples is then determined by comparing the O.D. of the samples to the standard curve.
 Intra-assay Precision    Intra-assay CV (%) is less than 15%.
 Inter-assay Precision    Inter-assay CV (%) is less than 15%. [CV(%) = SD/mean 100]

Preparation and Storage    Store all reagents at 2-8 degree C
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of NADHPOX elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for NADHPOX purchase    MBS024112 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Nicotinamide Adenine Dinucleotide Phosphate Oxidase Gp91Phox (NADHPOX) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.625ng/ml-20ng/ml in biological research samples containing NADHPOX, with an estimated sensitivity of 0.1ng/ml. The ELISA analytical biochemical technique of the MBS024112 kit is based on NADHPOX antibody-NADHPOX antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect NADHPOX antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, NADHPOX. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Tissue Homogenate, Feces and Urine. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

UniProt Comments for NADHPOX    NOX4: Constitutive NADPH oxidase which generates superoxide intracellularly upon formation of a complex with CYBA/p22phox. Regulates signaling cascades probably through phosphatases inhibition. May function as an oxygen sensor regulating the KCNK3/TASK-1 potassium channel and HIF1A activity. May regulate insulin signaling cascade. May play a role in apoptosis, bone resorption and lipolysaccharide-mediated activation of NFKB. May produce superoxide in the nucleus and play a role in regulating gene expression upon cell stimulation. Isoform 3 is not functional. Isoform 4 displays an increased activity. Isoform 5 and isoform 6 display reduced activity. 7 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 1.6.3.-; Membrane protein, integral; Cell cycle regulation; Oxidoreductase; Nucleolus; Membrane protein, multi-pass

Cellular Component: focal adhesion; mitochondrion; membrane; perinuclear region of cytoplasm; endoplasmic reticulum; apical plasma membrane; integral to membrane; plasma membrane; stress fiber; cell junction; NADPH oxidase complex
