Preparation and Storage    The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
ISO Certification    Manufactured in an ISO 9001:2015 and ISO 13485:2016 Certified Laboratory.
Supply Chain Verification    Manufactured in a lab with traceable raw materials manufactured on site. Coordinated product portfolio of antibodies, pairs, conjugates, recombinant proteins, and immunoassay materials available. Please inquire.
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of PON1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PON1 purchase    MBS2022720 is a ready-to-use microwell, strip plate Quantitative Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Paraoxonase 1 (PON1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 3.12-200ng/mL in biological research samples containing PON1, with an estimated sensitivity of 1.27ng/mL. The ELISA analytical biochemical technique of the MBS2022720 kit is based on PON1 antibody-PON1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PON1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PON1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for PON1 elisa kit

Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of PON1 in human serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to PON1. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to PON1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain PON1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm 10nm. The concentration of PON1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Summary for PON1    The enzyme encoded by this gene is an arylesterase that mainly hydrolyzes paroxon to produce p-nitrophenol. Paroxon is an organophosphorus anticholinesterase compound that is produced in vivo by oxidation of the insecticide parathion. Polymorphisms in this gene are a risk factor in coronary artery disease. The gene is found in a cluster of three related paraoxonase genes at 7q21.3. [provided by RefSeq, Oct 2008]

UniProt Comments for PON1    PON1: Hydrolyzes the toxic metabolites of a variety of organophosphorus insecticides. Capable of hydrolyzing a broad spectrum of organophosphate substrates and lactones, and a number of aromatic carboxylic acid esters. Mediates an enzymatic protection of low density lipoproteins against oxidative modification and the consequent series of events leading to atheroma formation. Genetic variation in PON1 is associated with susceptibility to microvascular complications of diabetes type 5 (MVCD5). These are pathological conditions that develop in numerous tissues and organs as a consequence of diabetes mellitus. They include diabetic retinopathy, diabetic nephropathy leading to end-stage renal disease, and diabetic neuropathy. Diabetic retinopathy remains the major cause of new- onset blindness among diabetic adults. It is characterized by vascular permeability and increased tissue ischemia and angiogenesis. Homozygosity for the Leu-54 allele is strongly associated with the development of retinal disease in diabetic patients. Belongs to the paraoxonase family.
