Intended Uses    This ELISA kit is intended for laboratory in vitro research use only, not for drug, household or other use! The Stop Solution changes the color from blue to yellow and the intensity of the color is measured at 450 nm by a spectrophotometer. In order to measure the concentration of Human PON in the sample, this ELISA kit includes a set of calibration standards. The calibration standards are assayed at the same time as the samples and allow the operator to produce a standard curve of Optical Density versus Human PON concentration. The concentration of Human PON in the samples is then determined by comparing the O.D. of the samples to the standard curve.
 Intra-assay Precision    Intra-assay CV (%) are less than 15%.
 Inter-assay Precision    Inter-assay CV (%) are less than 15%.

Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of PON elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PON purchase    MBS041476 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Paraoxonase (PON) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 62.5 U/L - 2000 U/L in biological research samples containing PON, with an estimated sensitivity of 10 U/L. The ELISA analytical biochemical technique of the MBS041476 kit is based on PON antibody-PON antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PON antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PON. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Tissue Homogenate, Feces, Urine and Body Fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for PON elisa kit

Introduction: ELISA is a simple and highly sensitive method of analysis that allows for simultaneous and rapid quantification of a large number of samples. The assay is based on the specific recognition of the target compound (analyte/antigen) by antibodies which bind to the compound. The antigen-antibody complex is detected and measured with the aid of an enzyme-labeled antibody or antigen. Upon addition of a non-colored reagent, the enzyme produces a color reaction where the color intensity is directly or inversely proportional to the concentration of the analyte in the sample.

NCBI Summary for PON    The enzyme encoded by this gene is an arylesterase that mainly hydrolyzes paroxon to produce p-nitrophenol. Paroxon is an organophosphorus anticholinesterase compound that is produced in vivo by oxidation of the insecticide parathion. Polymorphisms in this gene are a risk factor in coronary artery disease. The gene is found in a cluster of three related paraoxonase genes at 7q21.3. [provided by RefSeq, Oct 2008]

UniProt Comments for PON    PON1: Hydrolyzes the toxic metabolites of a variety of organophosphorus insecticides. Capable of hydrolyzing a broad spectrum of organophosphate substrates and lactones, and a number of aromatic carboxylic acid esters. Mediates an enzymatic protection of low density lipoproteins against oxidative modification and the consequent series of events leading to atheroma formation. Genetic variation in PON1 is associated with susceptibility to microvascular complications of diabetes type 5 (MVCD5). These are pathological conditions that develop in numerous tissues and organs as a consequence of diabetes mellitus. They include diabetic retinopathy, diabetic nephropathy leading to end-stage renal disease, and diabetic neuropathy. Diabetic retinopathy remains the major cause of new- onset blindness among diabetic adults. It is characterized by vascular permeability and increased tissue ischemia and angiogenesis. Homozygosity for the Leu-54 allele is strongly associated with the development of retinal disease in diabetic patients. Belongs to the paraoxonase family.

Protein type: Phosphatase (non-protein); Motility/polarity/chemotaxis; EC 3.1.1.81; Lipid-binding; Secreted; EC 3.1.1.2; Secreted, signal peptide; EC 3.1.8.1; Hydrolase

Chromosomal Location of Human Ortholog: 7q21.3

Cellular Component: extracellular space; intracellular membrane-bound organelle; extracellular region

Molecular Function: protein homodimerization activity; arylesterase activity; phospholipid binding; calcium ion binding; aryldialkylphosphatase activity

Biological Process: response to nutrient levels; response to external stimulus; dephosphorylation; organophosphate catabolic process; response to toxin; positive regulation of transporter activity; carboxylic acid catabolic process; positive regulation of binding; aromatic compound catabolic process; phosphatidylcholine metabolic process
