Intended Uses    This Quantitative Sandwich ELISA kit is only for in vitro research use only, not for drug, household, therapeutic or diagnostic applications! It is intended to be determinated PERK concentrations in Human serum, plasma and other body fluids. Using Purified Human PERK antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add PERK and PERK antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of PERK in the samples is then determined by comparing the O.D. of the samples to the standard curve.
 Intra-assay Precision    Intra-assay CV (%) is less than 15%.
 Inter-assay Precision    Inter-assay CV (%) is less than 15%. [CV(%) = SD/mean x100]

Preparation and Storage    Store all reagents at 2-8 degree C
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of PERK elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PERK purchase    MBS014568 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Phospho-Exiracellular Signal-Regulated Kinase (PERK) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 125 pg/ml 4000 pg/ml in biological research samples containing PERK, with an estimated sensitivity of 10 pg/ml. The ELISA analytical biochemical technique of the MBS014568 kit is based on PERK antibody-PERK antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PERK antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PERK. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Tissue Homogenate, Feces, Urine and Body Fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

NCBI Summary for PERK    The protein encoded by this gene is a member of the PP2C family of Ser/Thr protein phosphatases. PP2C family members are known to be negative regulators of cell stress response pathways. This phosphatase dephosphorylates, and negatively regulates the activities of, MAP kinases and MAP kinase kinases. It has been shown to inhibit the activation of p38 and JNK kinase cascades induced by environmental stresses. This phosphatase can also dephosphorylate cyclin-dependent kinases, and thus may be involved in cell cycle control. Overexpression of this phosphatase is reported to activate the expression of the tumor suppressor gene TP53/p53, which leads to G2/M cell cycle arrest and apoptosis. Three alternatively spliced transcript variants encoding distinct isoforms have been described. [provided by RefSeq, Jul 2008]

UniProt Comments for PERK    PPPM1A: a Ser/Thr protein phosphatase of the PP2C family. Inhibits the activation of p38 and JNK kinase cascades induced by environmental stresses. Can also dephosphorylate cyclin-dependent kinases and may play a role in cell cycle control. Overexpression of this phosphatase activates the expression of p53, leading to G2/M cell cycle arrest and apoptosis.
