Specificity    This assay has high sensitivity and excellent specificity for detection of DHX58. No significant cross-reactivity or interference between DHX58 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between DHX58 and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of DHX58 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for DHX58 purchase    MBS7213202 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Probable ATP dependent RNA helicase DHX58 (DHX58) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 1.0-25ng/mL in biological research samples containing DHX58, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS7213202 kit is based on DHX58 antibody-DHX58 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect DHX58 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, DHX58. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for DHX58 elisa kit

Intended Uses: This DHX58 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human DHX58. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: DHX58 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DHX58 antibody and an DHX58-HRP conjugate. The assay sample and buffer are incubated together with DHX58-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DHX58 concentration since DHX58 from samples and DHX58-HRP conjugate compete for the anti-DHX58 antibody binding site. Since the number of sites is limited, as more sites are occupied by DHX58 from the sample, fewer sites are left to bind DHX58-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DHX58 concentration in each sample is interpolated from this standard curve.

UniProt Comments for DHX58    DHX58: Acts as a regulator of DDX58/RIG-I and IFIH1/MDA5 mediated antiviral signaling. Cannot initiate antiviral signaling as it lacks the CARD domain required for activating MAVS/IPS1- dependent signaling events. Can have both negative and positive regulatory functions related to DDX58/RIG-I and IFIH1/MDA5 signaling and this role in regulating signaling may be complex and could probably depend on characteristics of the infecting virus or target cells, or both. Its inhibitory action on DDX58/RIG-I signaling may involve the following mechanisms: competition with DDX58/RIG-I for binding to the viral RNA, binding to DDX58/RIG-I and inhibiting its dimerization and interaction with MAVS/IPS1, competing with IKBKE in its binding to MAVS/IPS1 thereby inhibiting activation of interferon regulatory factor 3 (IRF3). Its positive regulatory role may involve unwinding or stripping nucleoproteins of viral RNA thereby facilitating their recognition by DDX58/RIG-I and IFIH1/MDA5. Involved in the innate immune response to various RNA viruses and some DNA viruses such as poxviruses, and also to the bacterial pathogen Listeria monocytogenes. Can bind both ssRNA and dsRNA, with a higher affinity for dsRNA. Shows a preference to 5'-triphosphorylated RNA, although it can recognize RNA lacking a 5'-triphosphate. Belongs to the helicase family. RLR subfamily.
