Specificity    This assay has high sensitivity and excellent specificity for detection of PTPRN. No significant cross-reactivity or interference between PTPRN and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between PTPRN and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of PTPRN elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PTPRN purchase    MBS731293 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Protein Tyrosine Phosphatase Receptor Type N (PTPRN) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 2.5-50ng/mL in biological research samples containing PTPRN, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS731293 kit is based on PTPRN antibody-PTPRN antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PTPRN antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PTPRN. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for PTPRN elisa kit

Intended Uses: This PTPRN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human PTPRN. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay||PTPRN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PTPRN antibody and an PTPRN-HRP conjugate. The assay sample and buffer are incubated together with PTPRN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PTPRN concentration since PTPRN from samples and PTPRN-HRP conjugate compete for the anti-PTPRN antibody binding site. Since the number of sites is limited, as more sites are occupied by PTPRN from the sample, fewer sites are left to bind PTPRN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PTPRN concentration in each sample is interpolated from this standard curve.

NCBI Summary for PTPRN    The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP possesses an extracellular region, a single transmembrane region, and a single catalytic domain, and thus represents a receptor-type PTP. This PTP was found to be an autoantigen that is reactive with insulin-dependent diabetes mellitus (IDDM) patient sera, and thus may be a potential target of autoimmunity in diabetes mellitus. Alternate splicing results in multiple transcript variants.[provided by RefSeq, Dec 2010]

UniProt Comments for PTPRN    PTPRN: Implicated in neuroendocrine secretory processes. May be involved in processes specific for neurosecretory granules, such as their biogenesis, trafficking or regulated exocytosis or may have a general role in neuroendocrine functions. Seems to lack intrinsic enzyme activity. May play a role in the regulation of secretory granules via its interaction with SNTB2. Belongs to the protein-tyrosine phosphatase family. Receptor class 8 subfamily. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Motility/polarity/chemotaxis; Membrane protein, integral; Receptor protein phosphatase, tyrosine

Chromosomal Location of Human Ortholog: 2q35-q36.1

Cellular Component: integral to membrane

Molecular Function: protein binding; GTPase binding; spectrin binding; protein tyrosine phosphatase activity

Biological Process: response to reactive oxygen species; response to cAMP; response to estrogen stimulus; insulin secretion; cytokine and chemokine mediated signaling pathway; response to glucose stimulus; protein amino acid dephosphorylation; response to insulin stimulus
