The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 18.75 pg/ml-1200 pg/ml in biological research samples containing RORA, with an estimated sensitivity of 4.68 pg/ml. The ELISA analytical biochemical technique of the MBS910208 kit is based on RORA antibody-RORA antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect RORA antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, RORA. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates, Cell lysates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for RORA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any RORA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RORA is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RORA bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a member of the NR1 subfamily of nuclear hormone receptors. It can bind as a monomer or as a homodimer to hormone response elements upstream of several genes to enhance the expression of those genes. The encoded protein has been shown to interact with NM23-2, a nucleoside diphosphate kinase involved in organogenesis and differentiation, as well as with NM23-1, the product of a tumor metastasis suppressor candidate gene. Also, it has been shown to aid in the transcriptional regulation of some genes involved in circadian rhythm. Four transcript variants encoding different isoforms have been described for this gene.
