Specificity    This assay has high sensitivity and excellent specificity for detection of FRZB. No significant cross-reactivity or interference between FRZB and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between FRZB and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of FRZB3 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for FRZB3 purchase    MBS7238827 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Secreted frizzled related protein 3 (FRZB) (FRZB3) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 2.5-50ng/mL in biological research samples containing FRZB3, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS7238827 kit is based on FRZB3 antibody-FRZB3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect FRZB3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, FRZB3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for FRZB3 elisa kit

Intended Uses: This FRZB ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human FRZB. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: FRZB ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FRZB antibody and an FRZB-HRP conjugate. The assay sample and buffer are incubated together with FRZB-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FRZB concentration since FRZB from samples and FRZB-HRP conjugate compete for the anti-FRZB antibody binding site. Since the number of sites is limited, as more sites are occupied by FRZB from the sample, fewer sites are left to bind FRZB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FRZB concentration in each sample is interpolated from this standard curve.

NCBI Summary for FRZB3    The protein encoded by this gene is a secreted protein that is involved in the regulation of bone development. Defects in this gene are a cause of female-specific osteoarthritis (OA) susceptibility. [provided by RefSeq, Apr 2010]

UniProt Comments for FRZB3    FRZB: Soluble frizzled-related proteins (sFRPS) function as modulators of Wnt signaling through direct interaction with Wnts. They have a role in regulating cell growth and differentiation in specific cell types. SFRP3/FRZB appears to be involved in limb skeletogenesis. Antagonist of Wnt8 signaling. Regulates chondrocyte maturation and long bone development. Defects in FRZB are associated with susceptibility to osteoarthritis type 1(OS1). Osteoarthritis is a degenerative disease of the joints characterized by degradation of the hyaline articular cartilage and remodeling of the subchondral bone with sclerosis. Clinical symptoms include pain and joint stiffness often leading to significant disability and joint replacement. Belongs to the secreted frizzled-related protein (sFRP) family.
