Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay.
Other Notes    Small volumes of TNFbeta elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for TNFbeta purchase    MBS722941 is a ready-to-use microwell, strip plate Competitive or Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Tumor Necrosis Factor beta (TNFbeta) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TNFbeta, with an estimated sensitivity of 1.0 pg/mL. The ELISA analytical biochemical technique of the MBS722941 kit is based on TNFbeta antibody-TNFbeta antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TNFbeta antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TNFbeta. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for TNFbeta elisa kit

For samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate

INTENDED USE This TNF? ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human TNF?. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

PRINCIPLE OF THE ASSAY TNF? ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for TNF?. Standards or samples are then added to the microtiter plate wells and TNF? if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of TNF? present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for TNF? are added to each well to “sandwich” the TNF? immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain TNF? and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TNF? concentration in each sample is interpolated from this standard curve.

UniProt Comments for TNFbeta    LTA: Cytokine that in its homotrimeric form binds to TNFRSF1A/TNFR1, TNFRSF1B/TNFBR and TNFRSF14/HVEM. In its heterotrimeric form with LTB binds to TNFRSF3/LTBR. Lymphotoxin is produced by lymphocytes and cytotoxic for a wide range of tumor cells in vitro and in vivo. Genetic variations in LTA are a cause of susceptibility psoriatic arthritis (PSORAS). PSORAS is an inflammatory, seronegative arthritis associated with psoriasis. It is a heterogeneous disorder ranging from a mild, non-destructive disease to a severe, progressive, erosive arthropathy. Five types of psoriatic arthritis have been defined: asymmetrical oligoarthritis characterized by primary involvement of the small joints of the fingers or toes; asymmetrical arthritis which involves the joints of the extremities; symmetrical polyarthritis characterized by a rheumatoidlike pattern that can involve hands, wrists, ankles, and feet; arthritis mutilans, which is a rare but deforming and destructive condition; arthritis of the sacroiliac joints and spine (psoriatic spondylitis). Belongs to the tumor necrosis factor family.

Protein type: Secreted; Cytokine; Secreted, signal peptide; Apoptosis

Cellular Component: extracellular space; membrane; extracellular region

Molecular Function: protein binding; cytokine activity; tumor necrosis factor receptor binding

Biological Process: positive regulation of humoral immune response mediated by circulating immunoglobulin; transformed cell apoptosis; positive regulation of apoptosis; lymph node development; humoral immune response; cell proliferation; positive regulation of interferon-gamma production; defense response to Gram-positive bacterium; negative regulation of fibroblast proliferation; positive regulation of cell proliferation; immune response; positive regulation of chronic inflammatory response to antigenic stimulus; inflammatory response; cellular process
