Specificity    This assay has high sensitivity and excellent specificity for detection of VPS-29. No significant cross-reactivity or interference between VPS-29 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between VPS-29 and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of VPS29 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for VPS29 purchase    MBS7238898 is a ready-to-use microwell, strip plate Competitive (Quantitative) ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Vacuolar protein sorting associated protein 29 (VPS29) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing VPS29, with an estimated sensitivity of 1.0 pg/mL. The ELISA analytical biochemical technique of the MBS7238898 kit is based on VPS29 antibody-VPS29 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect VPS29 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, VPS29. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for VPS29 elisa kit

Intended Uses: This VPS-29 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human VPS-29. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: VPS-29 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-VPS-29 antibody and an VPS-29-HRP conjugate. The assay sample and buffer are incubated together with VPS-29-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the VPS-29 concentration since VPS-29 from samples and VPS-29-HRP conjugate compete for the anti-VPS-29 antibody binding site. Since the number of sites is limited, as more sites are occupied by VPS-29 from the sample, fewer sites are left to bind VPS-29-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The VPS-29 concentration in each sample is interpolated from this standard curve.

NCBI Summary for VPS29    This gene belongs to a group of vacuolar protein sorting (VPS) genes that, when functionally impaired, disrupt the efficient delivery of vacuolar hydrolases. The protein encoded by this gene is a component of a large multimeric complex, termed the retromer complex, which is involved in retrograde transport of proteins from endosomes to the trans-Golgi network. This VPS protein may be involved in the formation of the inner shell of the retromer coat for retrograde vesicles leaving the prevacuolar compartment. Alternative splice variants encoding different isoforms and representing non-protein coding transcripts have been found for this gene. [provided by RefSeq, Aug 2013]

UniProt Comments for VPS29    VPS29: Essential component of the retromer complex, a complex required to retrieve lysosomal enzyme receptors (IGF2R and M6PR) from endosomes to the trans-Golgi network. Also required to regulate transcytosis of the polymeric immunoglobulin receptor (pIgR-pIgA). Has low protein phosphatase activity towards a serine-phosphorylated peptide derived from IGF2R (in vitro). Belongs to the VPS29 family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Protein phosphatase, Ser/Thr (non-receptor); Vesicle; EC 3.1.3.3

Chromosomal Location of Human Ortholog: 12q24

Cellular Component: intracellular membrane-bound organelle; retromer complex; cytoplasm; late endosome; early endosome; endosome membrane; cytosol; endosome

Molecular Function: protein binding; zinc ion binding; protein transporter activity

Biological Process: intracellular protein transport; retrograde transport, endosome to Golgi

Research Articles on VPS29    1. Conclusion is that VPS29 is a metal ion-independent, rigid scaffolding domain, which is essential but not sufficient for incorporation of retromer into functional endosomal transport assemblies.
