   The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Wingless Type MMTV Integration Site Family, Member 5A (WNT5A). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Wingless Type MMTV Integration Site Family, Member 5A (WNT5A). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Wingless Type MMTV Integration Site Family, Member 5A (WNT5A), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Wingless Type MMTV Integration Site Family, Member 5A (WNT5A) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

 NCBI Summary for WNT5A    The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene encodes a member of the WNT family that signals through both the canonical and non-canonical WNT pathways. This protein is a ligand for the seven transmembrane receptor frizzled-5 and the tyrosine kinase orphan receptor 2. This protein plays an essential role in regulating developmental pathways during embryogenesis. This protein may also play a role in oncogenesis.
