The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 1.25 pg/ml -80 pg/ml in biological research samples containing AVP, with an estimated sensitivity of <0.312 pg/ml. The ELISA analytical biochemical technique of the MBS700417 kit is based on AVP antibody-AVP antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect AVP antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, AVP. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as serum, plasma, tissue homogenates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

   Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ADH has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ADH present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADH is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADH bound in the initial step. The color development is stopped and the intensity of the color is measured.

 NCBI Summary for AVP    This gene encodes a precursor protein consisting of arginine vasopressin and two associated proteins, neurophysin 2 and a glycopeptide, copeptin. Arginine vasopressin is a posterior pituitary hormone which is synthesized in the supraoptic nucleus and paraventricular nucleus of the hypothalamus. Along with its carrier protein, neurophysin 2, it is packaged into neurosecretory vesicles and transported axonally to the nerve endings in the neurohypophysis where it is either stored or secreted into the bloodstream. The precursor is thought to be activated while it is being transported along the axon to the posterior pituitary. Arginine vasopressin acts as a growth factor by enhancing pH regulation through acid-base transport systems. It has a direct antidiuretic action on the kidney, and also causes vasoconstriction of the peripheral vessels. This hormone can contract smooth muscle during parturition and lactation. It is also involved in cognition, tolerance, adaptation and complex sexual and maternal behaviour, as well as in the regulation of water excretion and cardiovascular functions. Mutations in this gene cause autosomal dominant neurohypophyseal diabetes insipidus (ADNDI). [provided by RefSeq, Mar 2010]
