 Samples    Serum, plasma, tissue homogenates
 Assay Type    Sandwich
 Detection Range    12.5 pg/ml-800 pg/ml.
Sensitivity    The minimum detectable dose of human Casp-1 is typically less than 3.1 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision    Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision    Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

ISO Certification    Manufactured in an ISO 9001:2008 Certified Laboratory.
Other Notes    Small volumes of CASP1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for CASP1 purchase    MBS701433 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the caspase 1, apoptosis-related cysteine peptidase (interleukin 1, beta, convertase) (CASP1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 12.5 pg/ml-800 pg/ml in biological research samples containing CASP1, with an estimated sensitivity of The minimum detectable dose of human Casp-1 is typically less than 3.1 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. The ELISA analytical biochemical technique of the MBS701433 kit is based on CASP1 antibody-CASP1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CASP1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CASP1. Appropriate sample types may include undiluted human body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for CASP1 elisa kit

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for Casp-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Casp-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Casp-1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Casp-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Summary for CASP1    This gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce 2 subunits, large and small, that dimerize to form the active enzyme. This gene was identified by its ability to proteolytically cleave and activate the inactive precursor of interleukin-1, a cytokine involved in the processes such as inflammation, septic shock, and wound healing. This gene has been shown to induce cell apoptosis and may function in various developmental stages. Studies of a similar gene in mouse suggest a role in the pathogenesis of Huntington disease. Alternative splicing results in transcript variants encoding distinct isoforms. [provided by RefSeq, Mar 2012]

UniProt Comments for CASP1    Function: Thiol protease that cleaves IL-1 beta between an Asp and an Ala, releasing the mature cytokine which is involved in a variety of inflammatory processes. Important for defense against pathogens. Cleaves and activates sterol regulatory element binding proteins (SREBPs). Can also promote apoptosis.2 PublicationsManual assertion based on experiment in:Ref.3

Catalytic activity: Strict requirement for an Asp residue at position P1 and has a preferred cleavage sequence of Tyr-Val-Ala-Asp-|-.

Enzyme regulation: Specifically inhibited by the cowpox virus Crma protein.
