Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for YKL-40 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any YKL-40 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for YKL-40 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of YKL-40 bound in the initial step. The color development is stopped and the intensity of the color is measured.

 NCBI Summary for CHI3L1    Chitinases catalyze the hydrolysis of chitin, which is an abundant glycopolymer found in insect exoskeletons and fungal cell walls. The glycoside hydrolase 18 family of chitinases includes eight human family members. This gene encodes a glycoprotein member of the glycosyl hydrolase 18 family. The protein lacks chitinase activity and is secreted by activated macrophages, chondrocytes, neutrophils and synovial cells.
