 Samples    Serum, plasma, tissue homogenates
 Assay Type    Sandwich
 Detection Range    0.156 ng/ml -10 ng/ml.
Sensitivity    The minimum detectable dose of human GAD65 and is typically less than 0.039 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision    Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision    Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Preparation and Storage    Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
ISO Certification    Manufactured in an ISO 9001:2008 Certified Laboratory.
Other Notes    Small volumes of GAD2 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for GAD2 purchase    MBS702132 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the glutamate decarboxylase 2 (pancreatic islets and brain, 65kDa) (GAD2) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards render a theoretical kit detection range of 0.156 ng/ml -10 ng/ml in biological research samples containing GAD2, with an estimated sensitivity of The minimum detectable dose of human GAD65 and is typically less than 0.039 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. The ELISA analytical biochemical technique of the MBS702132 kit is based on GAD2 antibody-GAD2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GAD2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GAD2. Appropriate sample types include undiluted human body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for GAD2 elisa kit

Princple of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for GAD65 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GAD65 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GAD65 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GAD65 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Summary for GAD2    This gene encodes one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantibody and an autoreactive T cell target in insulin-dependent diabetes. This gene may also play a role in the stiff man syndrome. Alternative splicing results in multiple transcript variants that encode the same protein. [provided by RefSeq, Oct 2008]
