Specificity    This assay has high sensitivity and excellent specificity for detection of GHRP-Ghrelin. No significant cross-reactivity or interference between GHRP-Ghrelin and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between GHRP-Ghrelin and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of GHRP-Ghrelin elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for GHRP-Ghrelin purchase    MBS733049 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the growth hormone releasing peptide-Ghrelin (GHRP-Ghrelin) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing GHRP-Ghrelin, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS733049 kit is based on GHRP-Ghrelin antibody-GHRP-Ghrelin antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GHRP-Ghrelin antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GHRP-Ghrelin. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for GHRP-Ghrelin elisa kit

Principle of the assay: GHRP-Ghrelin ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GHRP-Ghrelin antibody and an GHRP-Ghrelin-HRP conjugate. The assay sample and buffer are incubated together with GHRP-Ghrelin-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GHRP-Ghrelin concentration since GHRP-Ghrelin from samples and GHRP-Ghrelin-HRP conjugate compete for the anti-GHRP-Ghrelin antibody binding site. Since the number of sites is limited, as more sites are occupied by GHRP-Ghrelin from the sample, fewer sites are left to bind GHRP-Ghrelin-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GHRP-Ghrelin concentration in each sample is interpolated from this standard curve.

NCBI Summary for GHRP-Ghrelin    This gene encodes the ghrelin-obestatin preproprotein that is cleaved to yield two peptides, ghrelin and obestatin. Ghrelin is a powerful appetite stimulant and plays an important role in energy homeostasis. Its secretion is initiated when the stomach is empty, whereupon it binds to the growth hormone secretagogue receptor in the hypothalamus which results in the secretion of growth hormone (somatotropin). Ghrelin is thought to regulate multiple activities, including hunger, reward perception via the mesolimbic pathway, gastric acid secretion, gastrointestinal motility, and pancreatic glucose-stimulated insulin secretion. It was initially proposed that obestatin plays an opposing role to ghrelin by promoting satiety and thus decreasing food intake, but this action is still debated. Recent reports suggest multiple metabolic roles for obestatin, including regulating adipocyte function and glucose metabolism. Alternative splicing results in multiple transcript variants. In addition, antisense transcripts for this gene have been identified and may potentially regulate ghrelin-obestatin preproprotein expression. [provided by RefSeq, Nov 2014]
