The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 62.5 pg/ml-4000 pg/ml in biological research samples containing UCP2, with an estimated sensitivity of 15.6 pg/ml. The ELISA analytical biochemical technique of the MBS930004 kit is based on UCP2 antibody-UCP2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect UCP2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, UCP2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell lysates. Product Description specifically for UCP2 elisa kit

   Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for UCP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any UCP2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UCP2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UCP2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

 NCBI Summary for UCP2    Mitochondrial uncoupling proteins (UCP) are members of the larger family of mitochondrial anion carrier proteins (MACP). UCPs separate oxidative phosphorylation from ATP synthesis with energy dissipated as heat, also referred to as the mitochondrial proton leak. UCPs facilitate the transfer of anions from the inner to the outer mitochondrial membrane and the return transfer of protons from the outer to the inner mitochondrial membrane. They also reduce the mitochondrial membrane potential in mammalian cells. Tissue specificity occurs for the different UCPs and the exact methods of how UCPs transfer H+/OH- are not known. UCPs contain the three homologous protein domains of MACPs. This gene is expressed in many tissues, with the greatest expression in skeletal muscle. It is thought to play a role in nonshivering thermogenesis, obesity and diabetes. Chromosomal order is 5'-UCP3-UCP2-3'. [provided by RefSeq, Jul 2008]
