MBS2503876 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the PSA/KLK3, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.313-20ng/mL in biological research samples containing PSA/KLK3, with an estimated sensitivity of The minimum detectable dose of Monkey PSA/KLK3 is 0.188ng/mL (The sensitivity of this assay, or lowest detectable limit (LDL) was defined as the lowest protein concentration that could be differentiated from zero). The ELISA analytical biochemical technique of the MBS2503876 kit is based on PSA/KLK3 antibody-PSA/KLK3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PSA/KLK3 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PSA/KLK3. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma and other biological fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to PSA/KLK3. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for PSA/KLK3 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain PSA/KLK3, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of PSA/KLK3. You can calculate the concentration of PSA/KLK3 in the samples by comparing the OD of the samples to the standard curve.

NCBI Summary for PSA/KLK3
|
|
Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Its protein product is a protease present in seminal plasma. It is thought to function normally in the liquefaction of seminal coagulum, presumably by hydrolysis of the high molecular mass seminal vesicle protein. Serum level of this protein, called PSA in the clinical setting, is useful in the diagnosis and monitoring of prostatic carcinoma. Alternate splicing of this gene generates several transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

UniProt Comments for PSA/KLK3
|
|
PSA: Hydrolyzes semenogelin-1 thus leading to the liquefaction of the seminal coagulum. Forms heterodimer with SERPINA5. Inhibited by SERPINA5. Activity is strongly inhibited by Zn2+, 100 times more abundant in semen than in serum. This inhibition is relieved by exposure to semenogelins, which are avid zinc binders. Belongs to the peptidase S1 family. Kallikrein subfamily.
Protein type: EC 3.4.21.77; Secreted; Protease; Secreted, signal peptide
Chromosomal Location of Human Ortholog: 19q13.41
Cellular Component: extracellular region; nucleus
Molecular Function: protein binding; serine-type peptidase activity; serine-type endopeptidase activity
Biological Process: negative regulation of angiogenesis; cellular protein metabolic process; proteolysis
