The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 78.125-5000pg/mL in biological research samples containing SOD1, with an estimated sensitivity of 46.875pg/mL. The ELISA analytical biochemical technique of the MBS2504429 kit is based on SOD1 antibody-SOD1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SOD1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, SOD1.    Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Monkey SOD1 concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Competitive-ELISA as the method. The microtiter plate provided in this kit has been pre-coated with SOD1. During the reaction, SOD1 in the sample or standard competes with a fixed amount of SOD1 on the solid phase supporter for sites on the Biotinylated Detection Ab specific to SOD1. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of SOD1 in the samples is then determined by comparing the OD of the samples to the standard curve.

 NCBI Summary for SOD1    The protein encoded by this gene binds copper and zinc ions and is one of two isozymes responsible for destroying free superoxide radicals in the body. The encoded isozyme is a soluble cytoplasmic protein, acting as a homodimer to convert naturally-occuring but harmful superoxide radicals to molecular oxygen and hydrogen peroxide. The other isozyme is a mitochondrial protein. Mutations in this gene have been implicated as causes of familial amyotrophic lateral sclerosis.
