INTENDED USE This APO-B48 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse APO-B48. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

PRINCIPLE OF THE ASSAY APO-B48 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-APO-B48 antibody and an APO-B48-HRP conjugate. The assay sample and buffer are incubated together with APO-B48-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the APO-B48 concentration since APO-B48 from samples and APO-B48-HRP conjugate compete for the anti-APO-B48 antibody binding site. Since the number of sites is limited, as more sites are occupied by APO-B48 from the sample, fewer sites are left to bind APO-B48-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The APO-B48 concentration in each sample is interpolated from this standard curve.

 NCBI Summary for Apo-B48    This gene product is the main apolipoprotein of chylomicrons and low density lipoproteins. It occurs in plasma as two main isoforms, apoB-48 and apoB-100: the former is synthesized exclusively in the gut and the latter in the liver. The intestinal and the hepatic forms of apoB are encoded by a single gene from a single, very long mRNA. The two isoforms share a common N-terminal sequence. The shorter apoB-48 protein is produced after RNA editing of the apoB-100 transcript at residue 2180 (CAA->UAA), resulting in the creation of a stop codon, and early translation termination. Mutations in this gene or its regulatory region cause hypobetalipoproteinemia, normotriglyceridemic hypobetalipoproteinemia, and hypercholesterolemia due to ligand-defective apoB, diseases affecting plasma cholesterol and apoB levels.  UniProt Comments for Apo-B48    APOB: Apolipoprotein B is a major protein constituent of chylomicrons (apo B-48), LDL (apo B-100) and VLDL (apo B-100). Apo B-100 functions as a recognition signal for the cellular binding and internalization of LDL particles by the apoB/E receptor. Defects in APOB are a cause of familial hypobetalipoproteinemia type 1 (FHBL1). A disorder characterized by highly reduced plasma concentrations of low density lipoproteins, and dietary fat malabsorption. Clinical presentation may vary from no symptoms to severe gastrointestinal and neurological dysfunction similar to abetalipoproteinemia. Defects in APOB are a cause of familial ligand-defective apolipoprotein B-100 (FDB).
