Specificity    This assay has high sensitivity and excellent specificity for detection of CLEC4E. No significant cross-reactivity or interference between CLEC4E and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CLEC4E and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of CLEC4E elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for CLEC4E purchase    MBS7246171 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the C-type lectin domain family 4 member e, CLEC4E, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.5-10ng/mL in biological research samples containing CLEC4E, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS7246171 kit is based on CLEC4E antibody-CLEC4E antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CLEC4E antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CLEC4E. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for CLEC4E elisa kit

Principle of the assay: CLEC4E ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CLEC4E antibody and an CLEC4E-HRP conjugate. The assay sample and buffer are incubated together with CLEC4E-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLEC4E concentration since CLEC4E from samples and CLEC4E-HRP conjugate compete for the anti-CLEC4E antibody binding site. Since the number of sites is limited, as more sites are occupied by CLEC4E from the sample, fewer sites are left to bind CLEC4E-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLEC4E concentration in each sample is interpolated from this standard curve.

NCBI Summary for CLEC4E    This gene encodes a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. Members of this family share a common protein fold and have diverse functions, such as cell adhesion, cell-cell signalling, glycoprotein turnover, and roles in inflammation and immune response. The encoded type II transmembrane protein is a downstream target of CCAAT/enhancer binding protein (C/EBP), beta (CEBPB) and may play a role in inflammation. Alternative splice variants have been described but their full-length sequence has not been determined. This gene is closely linked to other CTL/CTLD superfamily members on chromosome 12p13 in the natural killer gene complex region. [provided by RefSeq, Jul 2008]

UniProt Comments for CLEC4E    CLEC4E: C-type lectin that functions as cell-surface receptor for a wide variety of ligands such as damaged cells, fungi and mycobacteria. Plays a role in the recognition of pathogenic fungi, such as Candida albicans. The detection of mycobacteria is via trehalose 6,6'-dimycolate (TDM), a cell wall glycolipid. Specifically recognizes alpha-mannose residues on pathogenic fungi of the genus Malassezia. Recognizes also SAP130, a nuclear protein, that is released by dead or dying cells. Transduces signals through an ITAM-containing adapter protein, Fc receptor gamma chain /FCER1G. Induces secretion of inflammatory cytokines through a pathway that depends on SYK, CARD9 and NF-kappa-B.

Protein type: Membrane protein, integral; Receptor, misc.

Chromosomal Location of Human Ortholog: 12p13.31

Cellular Component: integral to membrane

Molecular Function: receptor activity; carbohydrate binding

Biological Process: immune response; positive regulation of cytokine secretion
