The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CD81, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS7241650 kit is based on CD81 antibody-CD81 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CD81 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CD81. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for CD81 elisa kit

   Intended Uses: This CD81 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CD81. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: CD81 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CD81 antibody and an CD81-HRP conjugate. The assay sample and buffer are incubated together with CD81-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD81 concentration since CD81 from samples and CD81-HRP conjugate compete for the anti-CD81 antibody binding site. Since the number of sites is limited, as more sites are occupied by CD81 from the sample, fewer sites are left to bind CD81-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD81 concentration in each sample is interpolated from this standard curve.

Interacts with a 16-kDa Leu-13 protein to form a complex possibly involved in signal transduction. May act as the viral receptor for HCV. Defects in CD81 are the cause of immunodeficiency common variable type 6 (CVID6); also called antibody deficiency due to CD81 defect. CVID6 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B-cells is usually in the normal range, but can be low.
